Tests
were performed in 96 well plates but other multiwell plates would
be suitable too. C. elegans was used as test organism in all sizes
from adult ones to small juveniles as reproduction was observed
during the time range of image acquisition. To provide acceptable
living conditions plates were kept at room temperature (about
20 °C). For feeding a suspension of E. coli was provided.
Test media was used corresponding to the German standard DIN draft
of ecotoxicological nematode testing. Where dilution was necessary
tap water was used. Image acquisition was performed at a range
of different magnifications from 1 to 24 wells per image. Observation
time was in the range between 10 and 100 seconds.

Using
advanced image processing steps the moving organisms can be extracted
from each image of an image series acquired. This series of recognised
moving nematodes can be used as basis for detailed movement or
behavioural analysis if single nematodes should be analyzed in
detail.One example for this is shown on the following images.
